Abstract

Molecular Diagnosis and Typing of Avibacterium paragallinarum Strains Isolated from Chickens The aim of this study was isolation, identification, molecular diagnosis and typing of Avibacterium paragallinarum isolated from broiler, broiler breeder and layer flocks with suspect of a respiratory disease. For this purpose, a total of 500 infraorbital sinus swab samples from 95 farms were examined by both culture and PCR. The swap samples were cultivated on special growth media for isolation and incubated in suitable conditions. After incubation colonies showing typical macroscopic and microscopic morphology were purified and subjected to confirmatory biochemical testing for identification. After confirmed by PCR, Kume reference strains and field isolates of A. paragallinarum were tested by ERIC-PCR for molecular fingerprinting.A. paragallinarum was isolated from 3 (3,15 %) layer flocks of 95 flocks investigated. Therefore, A. paragallinarum isolation was accomplished only from layer flocks. Neither culture nor direct PCR detected any positive chicken amongst broiler breeder and broiler flocks. Direct PCR detected 30 chickens / 11 layer flocks as A. paragallinarum positive. Typing of Kume reference strains and A. paragallinarum isolates by ERIC-PCR revealed different ERIC patterns. ERIC patterns of isolates from same farm were found to be same.In conclusion, higher positive rate obtained by PCR compared with culture from chickens with suspect Infectious coryza, indicated the usefullness and applicability of PCR technique in routine diagnosis of A. paragallinarum. The finding that PCR and culture positive samples originated from only layer flocks showed the importance of the infection to be much more in layer chickens. Obtained results showed that to prevent from loses due to A. paragallinarum infections, control and prevention measures containing vaccinations must be taken accurately
